Biomaterial-driven kidney organoid maturation

چکیده
Differentiation of human-induced pluripotent stem cells (hiPSCs) toward kidney organoids is known to suffer from batch-to-batch differences, off-target populations, and skewered cellular compositions. Application synthetic hydrogels as a tool for hiPSC-differentiation may provide additional control over this variable process. This review discusses important material properties that affect organoid generation. We summarize adhesive cues materials, allow transduction the mechanical forces cell, how these signals directly Hippo-Canonical Wnt signaling morphogenic events. In addition, chemical strategies are discussed spatiotemporal presentation biochemical agents cell in material-dependent approach.

Paracrine signals regulate human liver organoid maturation from induced pluripotent stem cells.
A self-organizing organoid model provides a new approach to study the mechanism of human liver organogenesis. Previous animal models documented that simultaneous paracrine signaling and cell-to-cell surface contact regulate hepatocyte differentiation. To dissect the relative contributions of the paracrine effects, we first established a liver organoid using human induced pluripotent stem cells ...

Organoid systems leverage the self-organizing properties of stem cells to create diverse multicellular tissue proxies. Most organoid models only represent single or partial components of a tissue, and it is often difficult to control the cell type, organization, and cell-cell/cell-matrix interactions within these systems. Herein, we discuss basic approaches to generate stem cell-based organoids...

Three-dimensional (3D) organoids provide a new way to model various diseases, including cancer. We made use of recently developed kidney-organ-primordia tissue-engineering technologies to create novel renal organoids for cancer gene discovery. We then tested whether our novel assays can be used to examine kidney cancer development. First, we identified the transcriptomic profiles of quiescent e...

Three-dimensional (3D) stem cell differentiation cultures recently emerged as a novel model system for investigating human embryonic development and disease progression in vitro, complementing existing animal and two-dimensional (2D) cell culture models. Organoids, the 3D self-organizing structures derived from pluripotent or somatic stem cells, can recapitulate many aspects of structural organ...
